Nowadays auto-vascular transplantation is used widespreadly. Compared with artifical vascular, it has its superiority , but its late stenosis rate is still about 30-40% and attracts some vascular specialist?attention. In vascular surgery, how to prevent the transplanted blood vessel from stenosis has become the focus of research. It has been confirmed since 1987 that NO was a gas endogenous molecule of biological message. It was named "yearly molecule"by science magazine in 1992. NO is related to the tone of peripheral blood vessel, it also can diastole vessel, inhibit the excretion of VSMCs and its migration, and prevent monocyte and platelets from adhering to vascular endothelial cells. In vivi, NO is produced by NOS which can catalyze 5 L-Arginie. Usually NOS was divided into two subtypes, ie, cNOS and iNOS. Among them, the expression of cNOS mRNA is dependent on Ca2+ and Calmodulin (CaM), its concerntration is low stable within blood and tissue fluid. The transcription of INOS is by blood flow shearing force and induced by inflammatory factor such as TNF-α. IL-1. The expression of iNOS increases and maintains at a high level for a long time at the action of inducing factors. Some reports have showed that ET is an endogenous molecule which is antagonistic agent against the action of NO. Our study is to investigate the time at which the isolated rabbit femoral veins were transplanted, and NO within the transplanted vein increased and maintained at a high level for long time, which would hinder the hyperplasia of transplanted vascular intima maximally. Serium NO concerntration was measured by Griess method. First nitrate reductase reduced NO3 to NO2 , then NO2 reacted with Griess agent. The value of OD was representive of the level of NO. The method we used is more precise than others. 6 We evaluated ET concemtration by RIA and measured 1NOS mRNA by RT-PCR. Experimental results showed the best time for auto-vascular transplantation. It may provide theoretical evidence to clinical treatment in future. 1. Experimental material: Fifteen male Newzealand rabbits with thirty hind legs. Serium sample are from femoral vein (FV) of rabbit's hind legs. 2. Experimental group and establishment of animal model. Fifteen rabbits were divided into five groups, each had three rabbits (six lengs). Group A: auto-vascular transplanted instantaneously Group B: auto-vascular transplanted with 2-hour-delayed. Group C: auto-vascular transplanted with 4-hour-delayed. Group D: auto-vascular transplanted with 8-hour-delayed. Group E: auto-vascular transplanted with 24-hour-delayed.
