Pharmacodynamics Research Of Non-hemorrhagic Fibrinolytic Enzyme From Agkistrodon Acutus

Objective: To purified fibrinolytic enzyme from Agkistrodon acutus venom and study the thrombolytic effect, anticoagulant effect and safety test. Methods: To purified non-hemorrhagic fibrinolytic enzyme by filtration and anion-exchange chromatography; To determine fibrinolytic activity by fibrin plate; To calculate molecular weight by SDS-PAGE; Platelet aggregation assays were perfomed in platelet rich plasma by turbidimetry; To measure thrombolytic effect on pulmonary embolism, arterial thrombosis , venous thrombosis; anticoagulant effect on animals and hemolytic test, acute toxicity test, hypersinsitive test and hemorrhagic test. Results: The NHFLE molecular weight was estimated as 25KD without any hemolyzation, hypersensitivity and hemorrhagic activity. NI-IFLE has directly fibrinolytic activity and inhibited not platelet aggregation induced by ADP.
