AIM: The interaction between CD40 and CD40 Ligand and gelatinase A, B (MMP-2,MMP-9)play an impotant role in pathophsiological functions in cardiovascular disease. The recently studies demonstrated that adhesion factors paticipated in myocardial ischemia reperfusion injury(MI/R) but had little known about gelatinase in MI/R.In this study we assessed the expression of CD40 and CD40 ligand(CD40L) andgelatinase in myocardial ischemia reperfusion injury in atherosclerosis rats.METHODS: All studies were performed on atherosclerosis diet rats modal with myocardial ischemia reperfusion(MI/R) (AS , n=41) and compared with the normal diet rats group (N, n=l 7) .The normal diet rats included control group(NC,n=5), ischemia lOmin group(NIiomin,n=6 ),ischemia 30min group (Nl30min,n=6 );and AS groups included control group(HC, n=5), the ischemia 30min group(n=6),the ischemia reperfusion Imin (HRlmin), 5min(HR5min ), 10min(HR10min), 20min (HR2omin),30min (HR3omin) groups(each grou p n=6);the changes of CD40 and CD40L expression of perepheral blood by flow cytometry and immuhischemical were detected;the activities of gelatinase (gelatinase A and gelatinase B) were examined; SOD and MDA were observed in each groups.RESULT :The means fluorescence intensity(MFI) of CD4(K CD40L and gelatinase and MDA in ischemia groups were higher than those in control group,P<0.05; the MFI of CD40, CD40L and gelatinase and MDA of HC* HlSOmin are higher than those of NC> NI30min,but SOD of H^ HDOmin was lower than the NCs NDOmin ; CD4(K CD40L and gelatinase A(MMP-2) in HR5minN HR-io^n are higher than the HI3omin (P<0.05);the indexs were the highest in R5mjn and then decreased in Riomin, R20min , Rsomin ,R30min was lower than the other reperfusion groups,but it showed no differences with the control group(P>0.05), but gelatinase B(MMP-9) had no different in the period of reperfusion groups; There had relationship between CD40VCD40L and MMP-2 (P<0.01),the relation-coefficient is 0.873 and 0.825; The longer reperfusion ,the higher of MDA and the lower of SOD were detected ;CD40-CD40L are expressed on the cardiocyte membrane, gelatinase A (MMP-2)\gelatinase B(MMP-9) are expressed on the cardiocyte membrane and myocardial matrix; especially on the ischemia myocardial.CONCLUSION: 1 CD40,CD40L and gelatinase maybe participate the myocardial ischemia ;2 CD40,CD40L and MMP-2 maybe participate the early of MI/R, which CD40,CD40L have the relationship with MMP-2; 3 CD40-CD40I^ gelatinase and oxidative stressparticipate the mechanism of atherosclerosis.
