OBJECTIVE: To investigate the influence of DC on exeprimental diabetes mellitus induced by high-sucrose and high-fat diet and streptozotocin,to provide data for further study and clinical practice.METHODS: Female wistar rats were fed with a diet enriched with sucrose (10%,w/w),triglyceride (10%,w/w) and cholesterol (l%,w/w) to induce insulin resistance ,and after one month on the diet, STREPTOZOTOCIN (stz,30mg/kg) was injected intraperitoneally to induce hyperglycemia,developing a rat model that closely stimulates the metabolic abnormalities of human type 2 disbetes mellitus .All female rats were randomly divided into eight groups . The first three groups were treated with DC :higher dose group(20g/kg),middle dose group(10g/kg) and lower dose group(5g/kg);the fourth group was treated with dendrobium alone;the other four groups were administered as control(two negitive groups and two postive groups).Meanwhile ,anther two groups were administered as normal control ones.The experiment lasted two months. Results were evaluated by measursment of water and food intaking ,blood-glucose,total cholesterol,triglyceride, serum insulin, plasma glucagons, urine glucose, urine ketones, GSP and body weight.After the experiment,get the pancreas and kidney out to serve as pathologyspecimens,and observe the change of pathology.RESULTS:!. Compared with negative control group, the level of blood-glucose decreased significantly (p<0.001) in dendrobium-treated and DC-treated groups.The effect is even better than that of positive groups . 2.The content of trilyceride ,total cholesterol ,GSP decreased significantly in dendrobium-treated and DC-treated groups compared with that in negative control group(p<0.01,P<0.05),while the positive control groups did not have such the effect (p>0.05).3.Compared with those in negative control groups,the other rats in both DC-treated and positive groups have gained weight (p<0.05)and weakened capacity for eating and drinking(p<0.05) .However there was no difference in these groups (p>0.05).4.The level of insulin increased in lower dose groups and middle dose groups of DC(p<0.05), but decreased in higher dose groups (p<0.01). 5.There was no difference between drug-treated groups and control groups in level of glucagon(p>0.05).6.Compared with negative control groups,the level of urine glucose decreased(p<0.05) ,there was no difference in DC-treated groups and positive groups (p>0.05).7.DC could decrease the damage of STZ to pancreas ,and protect the tissure of pancreas.The effect is better than that in the positive groups. CONCLUSIONSrDC can decrease blood-glucose, cholesterol, GSP, urine-glucose,triglyceride,GSP, protect the tissue of pancreas,adjust the level of insulin, and better the symptom of diabetes. Its effect on improving complication of diabetes is better than that of western medicine.
