Prostaglandin E₁ During Angioplasty As Preventive Therapy For Coronary Restenosis

Object: To evaluate the effectiveness of prostaglandin E1 on collagen and smooth muscle cells proliferation and the levels of Angll and t-PA after angioplasty in a rabbit model.Methods: Thirty healthy male New Zealand White rabbits were divided into three groups: the control group (n=10), the low dose prostaglandin EI group (n=10), and the high dose prostaglandin E\ group (n=10). Conventional balloon angioplasty was done in the right carotid artery, and twenty-six rabbits survived the operation: 8 in control group, 9 in low dose prostaglandin E\ group, and 9 in high dose prostaglandin EI group. Rabbits in prostaglandin E1 groups were given prostaglandin E1 0.3ug/kg/d, 1.0ug/kg/d infusion for two weeks after angioplasty. Arterial bloods were taken 30 minutes before angioplasty and 3 days, 7 days, 10 days, 15 days after angioplasty for analyzing plasma Angll, t-PA. For histology, Masson's and HE staining were performed two weeks later. All the specimens were analyzed using a computerized imaging analysis system and assess collagen measurement and smooth muscle cells proliferation.Results: 1.
