Part IObjective: To study the expression and localization of Vascular Endothelial GrowthFactor(VEGF) in humal1 Endometrium during ordinary menstrual cycle and in thehuman decidua of early pregnancy. Method: Using immunohistochemical analysis(streptavidin-biotin peroxidase method), we observed the expression of VEGF inendometriu1n during tl1e proIiferative phase (7cases), secretory phase (l2cases) and inthe decidua during early pregnancy(l 1cases). The VEGF semiqtlantitative score wascalculated as follows: l (VEGF positive cell<20%), II (VEGF positive cell between20-40%), III(VEGF positive cell between 40-60%), IV(VEGF positive cell>60%).By statistics we regarded the I as VEGF negative, lI, llland lV as VEGF positive.Results: There was only 2 cases(28.6%), which VEGF were expressed as positiveduring the proliferative phase, during the secretory phase l2 cases(l00%), and in thedecidua of early pregnancy l0 cases(90.9%), There was a significant difference,when the three groups were compared (P<0.01). The same results were made, whenthe proliferative phase group were compared with the secretory phase group (P<0.0l).But there was no significant difference between the secretory phase group and theearly pregnancy group to>0.05). The immunohistochemica1 localization of VEGFwas stronger in glands cell than in interstitial cell. The expression of VEGF in theendometrial glands cell was higher tI1an in interstitial cell.The immunohistochemicallocalization of VEGF 'vas in cytoplasm. Thelevels of FSH. LH. T. E2. P. PRL in serum were measured for all object atthe third day and in secretory phase (l9-22. day ) during the menstrual cycle.Result8: The average age in each group were: 28.92i4.34, 29.50f3.62, 28.92l3.00.There was no significant difference for disparity in age ty>0.05) .But there was asignificant difference in the expression and of Vascular Endothelial Growth Factor insecretory endometrium among three groups to<0.0l). The positive rate between theunexplained infertility group and control group was significant differential (P<0.0l),but no significant difference between the unexplained infertility group and that ofusing HCG group. Serum E2, P Ievels during secretory phase was significantdifferential betWeen the unexplained infertility group and the control group (P<0.0l).The means of hormone levels in control group were higher than that in unexplainedinfertility group. There was a significan differnce fOund in the means of E2, P andE/P during secretory phase between the unexplained infertility group and HCG group.The E2 and P levels in IICG group were higher than that in the unexplained infertilitygroup, but E/P lower (P<0.0l). Using Spearman's correlation-analysis, the expressionof VEGF and its hormone level in the unexplained infertility group and control groupwas compared .The results was not differential. Conclution: The unexplainedinfertility might be associated with VEGF. It might be a cause of the unexplainedinfertility , when the levels of E2, P lower during the secrtory phase.Using HCG will be advantageous to treat the unexplained infertility.
