| OBJECTIVETo investigate the expression of MUC1 in endometrium of women and,to evaluate the biological effects of MUC1 on endometrium, the window of implantation and its relation with unexplained infertility.METHODS(1)Expression of MUC1 is detected cases proliferative phase endometrium of in 10 cases, earlier period of luteal phase endometrium of in 10 cases,mid-luteal phase endometrium of in 16 cases, advanced stage of luteal phase endometrium of in 11 cases, respectively,by immunohistochemical method(Polymer Detection System).(2)Expression of MUC1 and MUC1 mRNA is detected cases normal mid-luteal phase endometrium of in 18 cases and unexplained infertile implantation endometrium of 19 cases, respectively, Immunohistochemical method (Polymer Detection System) and in situ hybridization method.(3)Expression of MUC1 is detected cases mid-luteal phase endometrium of in 18 cases, normal early pregnant endometrium of in 18 cases,inevitable abortion early pregnancy endometrium of in 20 cases,respectively,by immunohistochemical method(Polymer Detection System).RESULTS(1)MUC1 was diffusely distributed in the cytoplasm of luminal and glandular epitheliums. The glandular immunostaining score was lower in the proliferative phase.In earlier period of luteal phase ,the score is increased to 195.32±43.25,there is difference between the two group(P<0.05).In mid-luteal phase,the HSCORE is highest.The HSCORE was higher in mid-luteal phase compared that with earlier period of luteal phase (305.96±36.52 vs 195.32±43.25, P<0. 05).The HSCORE was higher in mid-luteal phase compared that with advanced stage of luteal phase(305.96 ±36.52 vs 186.58±48.13, P<0. 05).(2)MUC1 was diffusely distributed in the cytoplasm of luminal and glandular epitheliums in the two groups,the normal group and the infertile group. The endometrial glandular HSCORE in infertile group was significantly lower than that of the normal persons(216.54±47.31 vs 310.84±43.69, P<0. 05).MUC1 mRNA was diffusely distributed in the cytoplasm of luminal and glandular epitheliums in the two groups.The endometrial glandular HSCORE in infertile group was significantly lower than that of normal persons(155.43±41.87 vs 254.62±38.26, P<0. 05).(3)MUC1 was diffusely distributed in the cytoplasm of luminal and glandular epitheliums in the three groups,the normal group, the early pregnant groupand the inevitable abortion group.The endometrial glandular HSCORE didn't differ in the three groups(310.84±43.69,298.54±33.25,296.78±37.69, P>0. 05).CONCLUSIONS(1)The immunostaining of MUC1 in the endometrium was positive. The positive partical was located in cytoplasm of luminal and glandular epitheliums.MUC1 expressed in endometrium changed by menstrual cycle,is highly expressed in endometrium of mid-luteal phase(the implantation window of endometrium).(2)MUC1 is expressed highly in mid-luteal phase(the implantation window). MUC1 is the signal of the endometrial receptivity. MUC1 maybe one of the important factors that effect on the early blastocyst implantation.(3)The expression of MUC1 and MUC1 mRNA in unexplained infertility patient is diminished. And that lead to endometrial receptivity impaired maybe the cause of infertility.(4)After blastocyst implantation,the expression of MUC1 to endometrium is as high as that in mid-luteal phase.It means that MUC1 does't influence the growth of the embryo after it implantd into endometrium. |
