Objective To explore the significance and role of T lymphocytes, interleukin-10 on photoallergic contact dermatitis.Methods Balb/c female mice were used to establish the model of PACD to CPZ and ear swellings were evaluated at the different time point after the challenge ;Recombinant mice IL-10 was intradermally or intraperitoneally injected into the PACD model , ear swellings were evaluated at 48h after the challenge and mononuclears on the area of mm2 in H.E section were amounted ; Anti-mouse CD4 and(or) CD8mAb were intraperitoneally injected into the PACD model ,ear swellings were evaluated at 48h after the challenge, mononuclears on the area of mm2 in H.E section were amounted and the levels of IFN-γ,IL-10 in serum were checked. Results ①The model of PACD to CPZ was successfully established ;Ear swellings were obvious between 24h and 72h and reached the peak at 48h. ②Ear swellings and mononuclears of the mice injected i.p, intradermally by recombinant mouse IL-10 were lower as compared with the positive control group. ③when mice were injected i.p by Anti-CD4mAb,the level of IL-10 was close to and the level of IFN-γ was higher than those of the negative control group, But the levels of both were lower than those of the group injected i.p by Anti-CD8mAb;However ear swellings and mononuclears of groups injected i.p by Anti-CD8mAb and by Anti-CD4mAb were close, But their levels of IFN-γ, IL-10 were lower than those of the positive control group;ln the group injected i.p by Anti-CD8, CD4mAb,the levels of IL-10 - IFN-γ and ear swellings and mononuclears were close to the negative control group.Conclusions ① The mice model of PACD to CPZ is in conformation to delayed-type hypersensitivity induced by T lymphocytes; ②In vivo interleukin-10 can suppress the afferent and efferent phase of PACD to CPZ in the mice model; ③In the model of PACD to CPZ,CD4+Th1 and CD8+T lymphocytes regulate the reaction positively by increasing the level of IFN-γ; However , CD4+Th2 lymphocytes regulate the reaction negatively by increasing the level of IL-10.
