Objective: To explore the mechanism of KangXin liquid (a prescription for reinforcing Kidney and Spleen, removing blood stasis) in retarding aging by investigating the effect of KangXin liquid on liver mitochondrial DNA(mtDNA) deletion in aged Balb/c mice.Methods: 1. The liver mtDNA of the young mice(6~8 weeks ), middle age mice(6~8 months) and aged Balb/c mice (12-14 months) were isolated respectively. By using polymerase chain react ion (PCR) technique to examine the fragment deletion of liver mtDNA.2. In our natural aging model, the mice were divided into two groups, normal aged controlled group giving 0.9% NS , KangXin liquid group giving KangXin liquid. After four months, the deletions of liver mtDNA were detected by using PCR and electrophoresis. Analysis of deletions was proceeded in two different-group mice.Results: 1. There were 0.63bp mtDNA deleted in liver mitochondrial of aged Balb/c mice, but same mtDNA deletions were not detected in liver mitochondrial of young and middle age mice. 2. Compared with aging controlled group, KangXin liquid could reduce the mitochondrial DNA deletions in liver mitochondrial of aged mice.Conclusion: 1. mtDNA deletions are possible and could accumulate with age. 2. KangXin liquid can obviously inhibit the increase of levelof liver mtDNA deletions to retard aging in aged mice. |
