[Objective] :To examin the Ki-67 LI,MVD,DNA cytometry and ploidy andthe expression of Cathepsin-D, hTERT protein in breast cancer, combining the expression of ER, PR,P53, C-erbB-2 and tumor size, histological grading;to make clear the relationship between the biomarkers to lymphnode metastasis and to provide the valuable information of lymphnode metastasis and prognosis in breast cancer.[Methods]:1. The methods of computer image analysis was used to investigate the Ki-67 LI, MVD, DNA cytometry and ploidy.2. Using immunnohistochemical method,the expression of Cathepsin-D, hTERT protein,Ki-67, anti-FVIII-RA was investigated in 90 breast cancer, 28 precancerous dieases and 25 breast benign dieases.3. Logistic stepwise regression method was used to analyze the influence of 10 clinicopathological factors linking to lymphnode metastasis.[Results]:1. Ki-67 LI,MVD, DNA cytometry and ploidy(U) are increasing with breast benign dieases , precancerous dieases and breast cancer, there was a statistical significance. 2. Ki-67 LI (Labeling Index) is higher in invasive duct carcinoma and lower in intraductal carcinoma(P<0.05), was positive correlation with expression of ER, PR(P<0. 05) lymphnode metastasis(P<0. 01 )and negativecorrelation with expression of C-erbB-2; DNA ploidy(U) and Cathepsin-D showed positive correlation with tumor size,histopathological, lymphnode metastasis and expression of ER, PR, and negative correlation with expression of C-erbB-2;MVD showed positive correlation with tumor size, histopathological lymphnode metastasis ( P<0.01 ) ; hTERT protein showed positive correlation with histopathological and expression of ER, PR, P53; 3. In 10 clinicopathological factors , the closely linked to lymphnode metatasis are tumor size, histopathological, MVD, Cathepsin-D. [Conclusions]:1. Ki-67 LI and DNA ploidy can inflect the cell proliferation activity, which indirectly forecast the prognosis.2. The expression of Cathepsin-D is closely linked to lymphnode metatasis and prognosis of breast cancer.3. MVD which reflects angiogenesis of breast cancer is a important factor to lymphnode metatasis.4. Tumor size and histological grading are highly linked to the lymphnode metatasis and prognosis of breast cancer.5. The methods of computer image analysis will provide convenient for the diagnosis and the judgement of lymphnode metatasis.6. The usage of several biomarkers will improve the lightness in forecasting the prognosis of breast cancer.
