Objectives To test reproductive and genetic toxicity induced by water-soluble contents of cigarette smoke(WSCCS), and explore the mechanism.Methods The Horn's test , the micronucleus test in bone marrow PCE test,chromosome aberration test in primary spermatocyte, sperm aberration test and single-cell gel electrophoresis were undertaken in mice exposed to WSCCS.Results LD50 of WSCCS was 56.2mg/kg, 95% confidence interval was (33.8-82.5) mg/kg, refering to the classify stardard of toxicant, the toxicity of WSCCS belonged to middling toxicity. In MN test, in the low, middle and high dose groups , the mean frequencies of micronucleus (4.0%, 6.2% , 10.2%) was higher significantly than that in the negative control (1.8%o), the results also showed the obvious dose-response-relationship between the frequencies of micronucleus and the dose of WSCCS. In the middle , high dose groups of WSCCS, the mean frequencies of sex chromosome univalent (5.2%, 6.4%), the mean frequencies of sperm demafority(31.0%, 45.2%), were higher than those in the negative control(0.9% in chromosome aberration, 15.4% in spermaberration ) . Significant difference was observed in the mean frequenecies of autosome chromosome , the frequencies of structural aberration between the high dose group and the negative control. In comet test, all the WSCCS groups could induce the breakage of DNA single strand and result in comet cells with tail, the percentage in all dose groups was higher significantly than that in the negative control(P<0.01), and damage increased with the dose increased.Conclusion WSCCS included a great variety harmful materials, acute toxicity was stronger in mice, refering to the classify stardard of toxicant, the toxicity of WSCCS belong to middling toxicity. The results suggested WSCCS had mutagenecity, male reproductive and genetic toxicity.
