Objective To study the effect of propofol or/and procaine on the expression of CD18 , oxygen radical production of phorbol 12-myristae 13-acetate(PMA) stimulated human neutrophils(PMN) in vitro and on interleukin-6(IL-6) production in endotoxin-stimulated human whole blood . Method Using model of neutrophils in vitro, neutrophils were randomly divided into nine groups: control group(PMN+PBS), injure group(PMN+PBS+PMAlOOng.mF1), three different levels of propofol groups(PMN+ PMAl00ng.ml-1+ Propofol 0.4/4/40ug. mV), three different levels of procaine groups (PMN+PMA100ng. ml-1+Procaine1.5/15/150ug. ml-1), compound group (PMN+PMA100ng. ml-1+Propofol 2ug. ml-1+Procaine8ug.ml-1).Additionally different levels of propofol or/and procaine were selected in order to exclude the effect of propofol or /and procaine on the function of PMN . CD18 expression stimulated by PMA in vitro were measured by flow cytometry, superoxide anion release of human neutrophils stimulated by PMA was monitored by determined of cytochrome C reduction in the presence or absence of superoxide dismutase (SOD), IL-6 production in endotoxin-stimulated human whole blood were measured by radioimmunoassay. result Propofol or/and procaine inhibited PMA-induced up-regulation of CD18,release of superoxide anion of human neutrophils in vitro in a concentration-dependent manner and the effect of propofol is more powerful than the effect of procaine . The effect of compound-group was close to clinical concentration of propofol. Propofol strengthen the release of IL-6 in the whole blood in a concentration-dependent manner,procaine inhibit the production of IL-6 and has no difference with differentlevels. The effect of compound-group on the release of IL-6 was close to clinical concentration of procaine. Conclusion propofol or/and procaine at a certain level can be helpful for attenuation of organism damage induced by human netrophils .
