| The lung cancer was the most commonly tumor and disease in the world, non-small cell lung cancer, NSCLC was about 80% on the whole lung cancer, and prognosis was very poor, NSCLC included adenocarcinoma , squanoma and big-cell non-polarizative cancer. The invasive growth and metastasis of carcinoma is the mostly reason of low-survival rate in NSCLC recently.Apoptosis abnormally was very important in the pathogenic of lots of carcinoma. Survivin was found IAP recently, effected on the foucs of all kinds of apoptosis. It was the most strong apoptosis inhibitor factor so far. It was found that high expression of survivn was related to metastasis and invade in many tumor cell,but little to NSCLC.Early invade and metastasis was the main reason that early-dignosis difficultly , defeated treatment and death about tumor . Tenascin(TN)was a special six-arm glucoprotein outer cell metrix ,but not expressed in mature tissue and high expression in the tumor tissue. It was reported in a lots of carcinoma , but not NSCLC.The article is explored that the expression of Survivin and Tenascin ( TN) and if or not that the coordinated function between the factors and occurring , invading , metastasis about NSCLC theoretically. It was given that bases that improvement the level of treatment , the prognosis and treatment by tumor inhitor.Materials and MethodsPatients and specimen:NSCLC specimens were selected from paraffin wax embedded specimens with full cilnicopathological data. 60 patients underwent operations in the liaon-ing tumor hospital ,the period between sep,2003 and feb,2004. There were 24 males and 36 females,age 37-76 year old ,with mean age 57.2 6.2. There were 60 cases of lung cancer tisses and 30 of aside cancer. Accoding to UICC 1997 new standard , stage I 9 cases , stage II 29 cases and III 22 cases. Pathological differentatiation: squarnoma 30 cases , adenocarcinoma 21 cases, bronchile-alve-olus carcinoma 9 cases. By histological , there were high and moderate 52 cases and low 8 cases. By lymph node stage : N033 cases,N17 cases,N220 cases.2. Main reagentsMice anti-human Survivin polyclonal antibody, Mice anti-human Tenascin polyclonal antibody ( Maixin Corp , Fu zhou , China ) .Methods of ExperimentEvery speciment from the paraffin wax blocks were cut into 3 sections in sequent, one do staining by HE , the other two by Survivin and Tenascin immuno-histochemical staining. DAB was basal staining. The first antibody : mice anti-human Survivin polyclonal antibody and mice anti-human TN monoclonal antibody by SP staining. Postive staining of breast cancer was used as a postive control . Negative control sections were prepared by substituting primary antibody with PBS buffer solution.Standard of judgementBy microscope, Survivin postive is expressing yellow at cytoplasm of the tumor cell . According to the amount of postive cell and the depth of staining , there were classified by four levels: < 10% postive cell(-) ,above 10% postive cell( + ) , above 60% ( +++ ) , between 10% and 60% (++ ).TN positive expression was brown piece or net extracellual matrix , sections analyzed META MORPH/DP10/BX41 automatic medical color photoimagic analyzer.
