Study Of Total Saponin Of Aralia Elata Seem Leaf Effect On Tumor Cell Apoptosis And Expression Of Tumor Suppressor Gene P53

Aralia elata (Miq.)Seem is a kind of plant belong to Aralia Linn, of Araliaceae. The total saponin of Aralia elata Seem leaf is extracted from the leaves of Aralia elata Seem. The elementary pharmacology researches found that the TSAS was related to the anti-tumor effect. The TSAS can kill the tumor cells of S180 and H22, it can inhibit the growth of tumor cells significantly .The TSAS can markedly prolong the lives of mice bearing H22. It is found that the TSAS is not harm to the genetic material and can antagonize the effect of the mutagenic agent. Then it can change the components of the serum protein and tumor tissue protein of mice bearing H22, the effect is related to quality. And it can enhance the function of nonspecific immunity. To find the molecule mechanism of the anti-tumor effect of TSAS, we investigate the TSAS effect on cytokine, mtp53 and apoptosis in this study.In this study, we observed the effect of total saponin of Aralia eleta Seem leaf on expression level of mtp53 by ICH ,serum TNF-α by ELISA and the tumor cell apoptosis index by FCM in order to find the mechanism from the relation of mtp53 expression and apoptosis.Result indicate that TSAE can increase the TNF-α content (P<0.05), decrease the mtp53 positive expression rate (P<0.05), induce the tumor cell to apoptosis, the apoptosis index is 17.85% .
