Objective: To study the effects of decoction Shenghua on the expression of VCAM-l,ICAM-l,PECAM-l,iNOS and their corresponding mRNA in vascular endothelial cells(VECs) in rat models with blood stasis syndrome(BSS) and to explore the mechanism which it treats the diseases with BSS.Methods: The rat models with BSS were induced by injection of Adrenine (Adr) and instigation of ice, rats were divided into five equal groups randomly: the control group, the model group, high dose of decoction Shenghua therapy group, middle dose of decoction Shenghua therapy group and low dose of decoction Shenghua therapy group. The expression of VCAM-1, ICAM-1, PECAM-l,iNOS and their corresponding mRNA in VECs was assayed by immunohistochemistry, image analysis and RT-PCR.Results: Immunohistochemistry and image analysis indicated: no significant difference of VCAM-l's expression was shown (P>0.05) between the model group and the control group but its expression was lower in the therapy group than that in the model group, significant difference was shown between the two groups (the high dose group vs. the model group, P<0.05; the middle or low dose group vs. the model group, P<0.01); ICAM-l's expression was higher in the model group than that in the control group, significant difference was shown (P<0.05) between the two groups but no significant difference was shown between the therapy groups and the model group(P>0.05); No significant difference of PECAM-1's expression was shown (P>0.05) between the model group and the control group ,nor between the therapy groups and the model group(P>0.05); No significant difference of iNOS's expression was shown (P>0.05) between the model group and the control group but its expression was lower in the therapy group than that in the model group, significantdifference was shown between the two groups (the high dose group vs. the model group, P<0.05 ; the middle or low dose group vs. the model group, P<0.01; the high dose group vs. the low group, P<0.05); RT-PCR half-quantitative analysis indicated: mRNA was expressed in all five groups, mRNA's expression showed that it was higher in the model group than that in the control group, VCAM-1, PECAM-1, iNOS mRNA's expression was lower in the therapy groups than that in the model group, VCAM-1, iNOS mRNA's expression increased with the dose's decrease between the therapy groups, which was dose-dependent. ICAM-1 mRNA's expression was higher in the therapy groups than that in the model group.Conclusions: Decoction Shenghua can downregulate VCAM-1, iNOS's expression and VCAM-1, PECAM-1. iNOS mRNA's expression in VECs in rat models with BSS, which is probably one of important mechanisms that it treats the diseases with BSS.
