To the clinical medicine, it is a great development that the acute myocar- dial infarct patients have been cured by coronary artery repatency, thromboclasis, precutaneous transluminal coronary angioplasty (PTCA) and other heart surgery in the last years. The myocardial tissue receives blood reperfusion again due to cardiovascular repatency or reconstructive operation, which can really obtain favorable therapeutic efficacy in multitudinous conditions. However, the blood reperfusion will aggravate myocardial tissue injury and cardic insufficiency sometimes, even lead to serious arrhythmia. Therefore, it has important theropy meaning to seek safe and active traditional Chinese medicine to treat myocardial ischemia rep- erfusion injury complication.Our country has extremely abundant resources of traditional Chinese medicine and its development perspective is capacious. Ixeris Sonchifolia Hance(ISH) is widely distributed in Jilin province, which has been exploited Diemailing Injection and used in clinic so far. Many investigations has been done for Diemailing Injection in our laboratory. Prophase research has proved that Diemailing Injection has the effect of anti-acute myocardial ischemia and proved its active elements are flavone and adenosine. Injection of Ixeris Sonchifolia Hance (IISH) is the lyophilized powder extracted from Ixeris Sonchifolia Hance. On the base of the primary technology processor of Diemailing, the production technology of the lyophilized powder is perfected and the contents of its active elements-flavone and adenosine are increased evidently. We investigated the effect of IISH on experimental myocardial ischemia-reperfusion injury in rats and determined many experimental parameters including the changes of myo- cardial infarct size(MIS), the activity of serum creatine kinase, lactate dehydrogenase and aspartic transaminase, blood viscosity, the content of malonydialhyde(MDA) and the activity of sup- eroxide dismutase(SOD) in serum and infarct myocardium, the levels of plasma thromboxane A2(TXA2) and protacyclin(PGI2) and the contents of free fatty acids(FFA) in serum and infarct myocardium. In the investigation we discussed the effect and mechanism of IISH on anti-myocardial ischemia reperfusion injury, which offered experiment foundation for the development of ISH.1.Methods1.1 Groups and dosages:120 male wistar rats(weight 250-300g) were divided randomly into six groups including sham operation group, ischemia rep- erfusion(IR) model group, DML group(5.0g crude drug/kg i.p) and IISH low dosage group(2.5g crude drug/kg i.p), middle dosage group(5.0g crude drug/kg i.p), high dosage group(10g crude drug/kg i.p). Rats of sham operation group and IR group were given with physiological saline (5ml/kg i.p).There are 20 rats in every group. All rats were given drug one time every morning in two days and given drug one time immediately again after built the myocardial ischemia-reperfusion model in the third day morning. 1.2 Methods and parameters:After anesthetized with aether, rats were faced upward fixed on the operation table. Open the chest between the left third and fourth rib. The heart was exposed. A 0 nylon suture attached to a fine needle was placed under the left anterior descending coronary artery and a fine emulsoid tube was placed between coronary artery and suture. Then coronary artery was ligated with slipknot immediately. The heart was return into the chest, the blood and gas were squeezed out and the chest was closed swiftly. The time opening the chest is no more than 30 seconds. The sham operation group placed the suture only but not ligate coronary artery. After coronary artery was ligated 60 minutes, rats of each group were opened the chest and exposed the heart again. The nylon suture was undid immediately and the heart was reperfused.
