Objective: 1.To study the correlation between the expression of CD44V6 protein and PCNA and Estrogen Receptor(ER) and Progesterone Receptor(PR) status in breast cancer. 2. To investigate the value of determining CD44V6 protein and PCNA and ER and PR status for prognosis in breast cancer. 3. To investigate the value of determining CD44V6 protein in clinical use of breast cancer. Methods: By S-P immunohistochemical staining, the expression of CD44V6 and PCNA and ER and PR were investigated in paraffin-embedded tissues. Samples were obtained from 36 patients who had been diagnosed with primary breast cancer. The patients are all female from 20 to 74 years old. We had followed-up the patients till the end of September, 2003. There are nine patients died in ten years. Results: In 35 cases of breast cancer, the positive expression of CD44V6 were 25 cases (71.4%), which of PCNA were 24 cases (68.6%), that of ER were 19 cases (54.3%), and that of PR were 21 cases(60.0%). The incidence of positive ER was 40.0% the positive CD44V6 group, and was 90.0% in the negative CD44V6 group (P<0.01). The incidence of positive in the positive PR was 68.0% in the positive CD44V6 group while there are 60.0% in the negative CD44V6 group (P>0.05). There are 14 CD44V6(+) cases in movable carcinoma, while 11 cases in 14 immovable carcinoma, and there is no difference between two groups(P>0.05).The expression of CD44V6 was the highest in the simplex breast cancer (75%) in the breast cancer pathological classification, while in the invasive duct cancer, it was 42.9%, and the other classification was too less to be statisticed. There was no significant relationship between the simplex breast cancer and the invasive duct breast cancer (P>0.05). CD44V6(+) were 16 cases in 23 cases of clinical stage â… -â…¡(69.6%) and 9 cases in 12 cases of clinical stage â…¢-â…£(75.0%) (P>0.05).There are 8 cases in 10 cases of lowest and middle differentiation stage, while14 cases in 22 cases in highest differentiation (P>0.05).In 21cases positive axillary lymph nodes patients, there are 19 cases (90.4%), and 8 cases in 16 cases negative axillary lymph nodes patients(P>0.05). In the 19 cases of the premenopausal females, CD44V6(+) was13 cases (68.4%)(P>0.05), while 12 cases in 16 cases of the other(P>0.05). CD44V6(+) patients who survives over five years or more than five years was 72.0%(18 VS 25), which was higher than that of those who lived less than 5 years (P>.05). The rate of positive CD44V6 patients who had positive axillary lymph nodes was 90.4% (19/21),which is lower than that of the patients who had negative axillary lymph nodes(88.2%,15/17).There was significant difference between the two groups(P<0.05). The incidence of positive ER in the positive PCNA was 72.3%(8/11). In the negative PCNA group while the positive ER are 23.1%(3/13) in the negative PCNA group (P>0.05);The incidence of positive PR in the positive PCNA was 70.1%(17/24). In the negative PCNA group while the positive PR are 36.3%(4/11) in the negative PCNA group (P>0.05);There are 12 PCNA(+) cases in 21 cases movable carcinoma, while 13 cases in 14 immovable carcinoma, and the is signifianft difference between two groups(P<0.05);There was no significant relationship between the simplex breast cancer and the invasive duct breast cancer (P>0.05); There are 14 PCNA(+) cases in 16 cases of lowest differentiation stage, while10 cases in 19 cases in highest and middle differentiation (P<0.05);In 20 cases positive axillary lymph nodes patients, there are 17 cases (85.0%), and 7 PCNA(+) cases in 16 cases negative axillary lymph nodes patients(P<0.05); PCNA(+) patients who survives over five years or more than five years was (15 VS 9), which was higher than that of those who lived less than 5 years (P<0.05).
