ObjectiveCholangiocarcinoma may metastasis via lymphatic, bile duct and nerve at early stage, infiltrating growth is its major feature of Pathology. Because most patients have not special clinical situation and most patients are late stage at first presentation, its local infiltration and remote metastasis lead to difficult radical operation. Survival rate is low and prognosis is poor after operation. Nowadays the pathological examination is difGcult to reflect malignancy level and trend of metastasis and trend of infiltration exactly. Sometimes as indivdvial patients have a long life after palliation opertation, it shows that prognosis of cholangiocarcinoma is related to bionomics. Now study of bionomics focus on seaching for special tumor antigen . The discovered special bile duct tumor antigen has low sensitivity and specificity, so it is difGcult to apply in clinic. Presently RCAS1, a new tumor antigen, is receptor - binding cancer antigen expressed on human uterine adenocarcinoma cell line( SiSo) , and it had been studied widely in uterine adenocarcinoma, carcinoma of gallbladder, small cell lung cancer, thyroid carcinoma et al . RCASl often express in infiltrating carcinomas, and high expression indicate a poor prognosis, but there is few dependability research between the antigen and cholangiocarcinoma.In this study, we will examine the expression of RCASl in cholangiocarcinoma by immunohistochemistry ( S - P). And by integrating clinical data, we will verify the expression of RCAS1, and their relationship with clinicopathologi-cal significance of cholangiocarcinoma . we will analyze bionomics of cholangiocarcinoma in order to supply theoric bases for comprehensive treatment.Methods1. Tissue specimens collection: 42 cholangiocarcinomas speciments as experiment between 1999 and 2003 in the First AfiBliated Hospital Of China Medical University were obtained. Among 10 normal bile duct tissues as control, ten is from transplant samples. All the samples were confirmed by H&E before experiment.2. Main agents; RCASl antibody(mouse monoclonal antibody for human 22 -1 -1) are from Santa Cruz company in USA. S - P agents are from Maixin Biological company.3. Methods; The 42 cholangiocarcinoma specimens and 10 normal bile duct tissue were stained by immunohistochemistry ( S - P) and the expressionof RCASl was clarified with microscope. Wilcoxon Mann - Whitney test was used with SPSS10. 0 soft ware.Results1. The expression of RCASl in cholangiocarcinoma was higer than that in normal bile duct tissues, there was significant difference between them ( Z = -3.102,P<0.05).2. Between male and female, there was no sigificiant difference in the expression of RCASl. Less than 60 ages and greater than or equal 60ages, there was no sigificiant difference in the expression of RCASl. There was no significant difference in sex and age ( P > 0.05 ).3. The expression of RCASl in well - differentiated,moderately differentiated, poorly differentiated cholangiocarcinoma was significant difference. The expression of RCAS1 in TNM was significant difference . There is significant difference among them( P <0.01).
