Expression And Analysis Of Cell Apoptosis Genes And Integrinβ3 In Endometriosis

Objective:We investigated expression of Bcl-2,Bax,p53 and integrinβ3 in endometrium from women with and without endometriosis in order to explore the role of them in endometriosis and provide a theoretical basis for clinical therapy. Methods:We collected ectopic endometrial tissue and eutopic endometrial tissue from women with and without endometriois .Cellular localization and expression of Bcl-2, Bax and integrinβ3 protein were evaluated by immunohistochemistry. Expression of p53mRNA were evaluated by insitu hybridization.apoptotic cell were detected using amodified terminal deixynucleotidyltransferasebiotin nick end-labeling method (TUNEL).The expression of integrin β3mRNA were deteced by the reverse transcription polymerase chain reaction (RT-PCR). Results: (1) The apoptosis level of endometriosis was significant lower than women without endometriosis(p<0.01).The apoptosis of ectopic endometrium was higher than eutopic endometrium of endometriosis.(2) In controls and in eutopic endometrium of EM,the expression of Bcl-2 in proliferation was higher than in secretory(p<0.05;p<0.05) .Each group in ectopic endometrium was not significant different.But the expression of Bcl-2 in both eutopic and ectopic endometrium of EM was higher than the controls.(3) In controls and in eutopic endometrium of EM,the expression of Bax in secretory was higher than in proliferation (p<0.05;p<0.05) . Each group in ectopic endometrium was not significant different.But the expression of Bax in both eutopic and ectopic endometrium of EM was lower than the controls.(4) The expression of p53mRNA in both eutopic and ectopic endometrium of EM was lower than the controls.(5) The expression of integrin ?3 in EM was lower than the controls(p<0.05). Conclusions: Abnomal expression of those apoptosis genes in endometrium of women with endometriosis may be the possible or part reason in the progression of endometriosis .one of the main reasons for endometriosis might be the obviously decreased expression of integrin ?3 subunit in endometrium that contribute to the resuce of uterin receptivity.
