| In North America and Europe,the incidence and mortality of prostatic carcinoma(Pca) is outstanding.Recently,some evidences indicates that the incidence of prostatic carcinoma(Pca) is growing. Its increasing rate of the incidence has taken the first place in Urological and procreation malignancy in China.Prostatic carcinoma is characterized with the latency and high incidence.Percent 25 of men over 70 years old in our country suffer from prostatic carcinoma,but only a few of them can become clinical cancer.In therapy,chemical drugs are difficult to enter into prostatic tissue.And more prostatic carcinoma have metabasised when patients have clinical symptom,so surgery has bad effects.Now endocrine therapy can only improve patient's and lessen the pain of the patients,however,it can't prolong survival time,which call for new means to treat prostatic carcinoma.The growth of prostatic carcinoma is correlated with the balance between the growth and death of cells.IAP is very important in carcinogenesis of prostatic carcinoma.Survivn is a member of IAP family that was discovered recently. Survivn can inhibit the apoptosis of cell and promote the development of cancer.Now The research on oncotherapy through targeting Survivin expression have developed quckly.The present study employed RT-PCR and immunohistochemistry toexplore the expression of Survivin in normal prostate tissue(NP) , benign prostatic hyperplasia(BPH) and prostatic carcinoma tissue (Pca),aiming to study the possible role of Survivin.Aim:To investigate the expression of Survivin in human normal prostate tissue(NP),benign prostatic hyperplasia(BPH) and prostatic carcinoma(Pca) and the relationship between Survivin and pathologic grading and clinical stages of prostatic carcinoma to lay the foundation for further therapeutic experiment.Methods: The expression of Survivin mRNA in human normal prostate tissue(NP) benign prostatic hyperplasia(BPH),and prostatic carcinoma(Pca) was examined by RT-PCR.The expression of Survivin in human normal prostate tissue(NP) benign prostatic hyperplasia(BPH),and prostatic carcinoma(Pca)was examined by SABC way.Results: 1.The result of RT-PCR: In specimens of normal prostate tissue(NP),there are 4 male expressions ,the male expression rate was 13.3% (4/30) .In specimens of BPH, the male expression rate was 26.0%(13/50). In specimens of prostatic carcinoma (Pca), the male expression rate was 72.7%(43/55),which was significantly different from normal prostate tissue (NP) and benign prostatic hyperplasia (BPH)(P<0.05). The expression of Survivin had significant correlation with the pathologic grading as well as clinical stages of prostatic carcinoma(P<0.05).The Survivin mRNA was expressed weakly in normal prostate tissues (NP) and benign prostatic hyperplasia (BPH) but was expressed strongly in prostatic carcinoma (Pca).
