Hohenbuehelia serotina named Dong mushroom, Yuan mushroom, is a famous kind of edible wild fungus in northeast region. Hohenbuehelia serotina's pharmacological effects are complicated and the water-soluble Huangmo polysaccharides (FI) is extracted from it. The anti-tumor effect of FI and the effect of synergism, decreasing toxicity were studied in this experiment. We also studied on its mechanism through tumor immunity. 1. Anti-tumor experiment in vivo The bearing tumor mouse model with Hepatoma-22 was established for anti-tumor experiment. FI was administered through i.p. and divided into 3 dose groups, which is 20mg/kg,40mg/kg and 80mg/kg. After 10 days treatment with FI, low, middle and high dose group inhibition rate (IR) in Hepatoma-22 is 27.9%, 39.8%, 51.2% respectively; It is a good activity on anti-liver cancer. 2. The side and toxic reaction of FI We measured the body weight variation, thymus index, spleen index, white blood cell (WBC) amount of tumor bearing mice to determine the side and toxic reaction of FI. The results show that FI has no significantly effect on all of above index (compared with control group P>0.05). It indicates that the side and toxic reaction of FI are safe and slight. 3. Synergism Experiment We observed the synergism that FI improved the effect of CTX through the transplanting tumor model of Hepatoma-22. The result showed that FI could significantly improve the effect of CTX with dosage 15mg/kg and 30mg/kg in Hepatoma-22 4. Decreasing Toxicity Experiment. The anti-tumor effect was observed while compared toxicity, which used CTX only with the toxicity which used CTX and FI through the transplantation tumor model of Hepatoma-22. The result showed that FI could decrease the toxicity of CTX in anti-tumor and increase the body weight, thymus index, spleen index and white blood cell (WBC) amount of tumor bearing mice. 5. The effect of FI on immune system 5.1 Increase the Weight of Immune Organs Result showed that FI 20mg/kg and 40mg/kg had the effect of increasing the weight of immune organs. Increasing the weight of thymus and spleen is 9.17~ 24.45 percent, 19.23~24.35 percent respectively. Those indicate that FI could stimulate lymphocyte proliferation and transformation. 5.2 Lymphocyte transformation experiment The incremental reaction of mouse lymphocyte was observed by the activity of spleen cell to mitosis ConA. Result showed that FI (50mg/kg, 100 mg/kg) had the effect of improving the lymphocyte transformation. 5.3 NK cell's activity To observe FI effect on bearing mouse NK cell's activity by MTT colorimetry technique in which Yac-Ⅰcell was used as targeting cell .The results showed that FI (20mg/kg, 80mg/kg) could improve the activity of NK cell significantly (compared with control group P<0.05). 5.4 The effect on IL-2 activity of bearing mouse The method of dependent cell strain was used to observe FI effect on IL-2 activity of bearing mouse. Result showed that FI 20mh/kg,40mg/kg,80mg/kg could improve the IL-2 activity of bearing mouse.
