Objective: The effects of exercise training and rosiglitazone treatment on the type 2 diabetes mellitus in OLETF rat were examined. Methods: The male OLETF rats (12 wk old) were divided into 4 groups, (control, rosiglitazone, exercise, rosiglitazone combined exercise group) and then chose male LETO rats for counterparts. The rosiglitazone group was fed with 3 mg/kg/day for 24 weeks. The rats of exercise group were investigated by running (12 m/min for 30 min/day) on a activate cage for 24 weeks. Intraperitoneal glucose tolerance test (IPGTT) was examined in all rats. The serum glucose and insulin levels, weight of body and epididymal adipose tissue were measured at 12, 24 and 36 weeks of age. The pancreas tissues were sampled and examined by immunohistochemical staining. The immunohistochemical staining was carried out by technique of avidine-biotin-peroxidase complex. Results: After 24 weeks, the body weight and weight of epididymal adipose tissue in exercise and exercise combined rosiglitazone group were significantly lower than control group (P<0.05). The area under curve of IPGTT in exercise, rosiglitazone (P< 0.05) and exercise combined rosiglitazone groups (P<0.001) were significantly decreased than control group. The plasma glucose to insulin ratio in 3 groups was higher than control group (P<0.05). Through the insulin immunocytochemistry assaying, the B cell in control group showed early apoptosis, as well as pathologic changes in nuclei and organelles. However, rosiglitazone, exercise, exercise combined rosiglitazone groups were significantly attenuated the B cell apoptosis. Conclusions: In this study, exercise could prevent weight gain and insulin resistance and the long-term exercise training was comparable to rosiglitazone treatment. Conclusively, combination therapy of exercise and rosiglitazone can potentates improving insulin resistance and preventing diabetes mellitus.
