Gastrointestinal motility is one of the most important in digestive system.It is coordinated several levels of the smooth muscles.The incidence of the gastrointestinal functional diseases rises gradually in China. Alimentary tract dyskinesis is very common.The systematic diseases are accompanied with the changes of Gastrointestinal motility besides ulser, chronic gastritis and esophagus inflammation.There are Western medicines such as maxolon, domperidone and cisapride.They are reliable and have conspicuous effects of promoting gastric motivation.But we should also consider the side effects of this kind of medicine.Treatments of gastric dyskinesis using Chinese herbs are accumulated plenty of experiences.The natural resoures of Chinese herbs are abundant and their side effects are much less,so people are interested in using Chinese herbs to treat gastric dyskinesis gradually.Therefore , this experiment employed the method of perfusing isolated gastric muscle strips to study the effects of monomers,decoction on contraction of isolated gastric muscle strips in guinea pigs and the possible mechanism involved.The experimental results were as follows:一, Effects of magnolol on contractile activity of isolated circular muscle strips of gastric body in guinea pigs.1. Magnolol (10-6 mol/L,10-5.5mol/L,10-5 mol/L,10-4.5 mol/L,10-4mol/L and 10-3.5mol/L) dose dependently decreased the mean contractile amplitude and the resting tension of circular muscle(CM) of gastric body. Magnolol (10-6 mol/L,10-5.5mol/L,10-5 molL,10-4.5 mol/L,10-4mol/L and 10-3.5mol/L) did not affect the contractile frequency of CM of gastric body.Propranolol (10-6mol/L), L-NNA (10-4mol/L), indomethacin (10-5mol/L) and phentolamine (10-6mol/L) did not inhibit the decreasing action of the mean contractile amplitude and the resting tension of CM of gastric body.2. CaCl2 (10-5mol/L,10-4.5mol/L,10-4mol/L,10-3.5mol/L,10-3mol/L,10-2.5mol/L and10-2mol/L) induced CM of gastric body dose dependently contract in calcium- free Krebs solution. Magnolol caused concentration-response curve to move right and decreased themaximum reaction.3. After CM of gastric body was equilibrated for 30 minutes in calcium-free Krebs solution,ACh(10"5mol/L) was added. ACh could induce a fast and temporal contraction(the contraction of phase I ).When this contraction reached to its maximum reaction, CaCl2(10"2mol/L) was added in the tissue chamber.The strip produced a lasting contraction(the contraction of phase II). Magnolol could significantly inhibit the contraction of phase II induced by CaCh.The results indicated that mognolol possessed inhibiting action on the contraction of CM of gastric body. The action of magnolol was possibly due to the inhibition of voltage-dependent Ca2+ channels(VDCCs). Magnolol did not inhibit the release of Ca2+ from sarcoplasm reticulum induced by ACh ,but the inflow of Ca2+ from the extracelluar.H, Effects of emodin on contractile activity of isolated circular muscle strips of gastric body in guinea pigs.1. Emodin (10"6mol/L,10"5 5 mol/L,10'5 mol/L and 10"4 5 mol/L) increased the mean contractile amplitude of CM of gastric body. Emodin (105 mol/L and 10"4 5 mol/L) increased the resting tension. Emodin (10"4'5 mol/L) quickened the contractile frequency of CM of gastric body.2. 4-DAMP(muscarinic3 cholinergic antagonist, 10'6mol/L), isoptin(L-type calcium channel blocker,10"5mol/L) partly blocked the increased mean contractile amplitude of emodin on CM of gastric body in guinea pigs.The experimental results showed emodin possessed exciting action on CM of gastric body in guinea pigs.The exciting action of emodin was partly mediated via cholinergic M3 receptor and L-type calcium channel.H .. Effects of areca peel on contractile activity of isolated circular muscle strips of gastric body in guinea pigs.1. Areca peel (3%,10%,30%,100%,200%)dose dependently increased the mean contractile amplitude of CM of gastric body in guinea pigs. Areca peel(100%, 200%) dose dependently raised the resting tension. Areca peel (10%, 30%, 100%, 200%)quickened the contractile frequency of CM of gastric body.2. 4-DAMP(muscarinic3 cholinergic antagonist) partly blocked the increased meancontractile amplitude of areca peel on CM of gastric body,The result showed that areca peel possessed exciting action on CM of gastric body in guinea pigs. The exciting action of areca peel is partly mediated via cholinergic M3 recepter,but not cholinergic M2 recepter.
