Methods: Using immunohistochemistry,reverse transcriptase-polymerase chain reaction (RT-PCR) and single strand conformation polymorphism (SSCP), the expression variation and gene mutation of nm23-H1 gene were detected in 30 NPC tissues and in 11 nasopharyngeal tissue of chronic pharyngitis. Results :①the rate of nm23-H1 protein express positive in 30 NPC tissue was 53.30%(16/30); but it was 90.91%(10/11) in nasopharyngeal tissue of chronic pharyngitis,the rate of nm23-H1 protein express positive in NPC which has lymphonodi cervicales metastasis was 40.91%(9/22); and it was 87.50%(7/8) in non-metastasis tissues. There were significant difference in the expression rate of nm23-H1 protein among the two kind of tissues(P<0.05);②The expression level of nm23-H1 protein in NPC tissues was not closely related to NPC patients' age and gender (P>0.05);③There was exist linear relationship between two transcripton of nm23-H1.There was significant difference in the expression rate of nm23-H1 mRNA among tissues of nasopharyngeal carcinoma and chronic pharyngitis tissues.It was the same of metastasis NPC tissues and non-metastasis tissues;④One of the nm23-H1 gene mutation was found in all the nasopharyngeal samples. Conclusion:①The rate of nm23-Hl protein express positive in the NPC tissues is very low;②The expression level of nm23-H1 in the NPC tissues is much lower than that in the chronic pharyngitis tissues, which indicates that the inactivation of nm23-H1 is associated with the pathogenesis of NPC;③The expression level of nm23-H1 in the metastasis NPC tissues is much lower than that in the non-metastasis NPC tissues,which indicates that the inactivation of nm23-H1 is associated with the metastasis of NPC;④The expression level of nm23-Hl protein in NPC tissues was not closely related to NPC patients' age, and gender in NPC tissues;⑤The gene mutation rate of nm23-H1 in the NPC tissues is very low. | | Keywords/Search Tags: | Nasopharyngeal carcinoma, nm23-H1, RT-PCR, PCR-SSCP | PDF Full Text Request | Related items |
