In this paper, the effective fraction was thought as the core of study on Chinese Traditional Compound Medicine(CTMC), and a new viewpoint was proposed, i. e, the group of effective components is the smallest molecule group obtained by modern separation means and still maintaining the therapeutic effect of original CTMC. In addition, it was thought that penetrating into the effective fraction would lay a foundation for realizing the deepening of CTCM theory from medicines compatibility to chemical components compatibility.On the bases of these viewpoints, a Chinese Traditional Compound Medicine— —Xianhe consisting of Agrimonia pilosa Ledeb., Hedyotis diffusa Willd, Solanum lyratum Thurb, and Glycyrrhiza uralensis Fisch, was systematical studied to separatethe effective fraction guided by the antitumor effective fractions--polysaccharide,tannins, flavonids, triterpenoids.The extraction process of Xianhe was optimized by the weight of dry extracts and the content of the sample.The purified determination method of polysaccharide in the alcohol extract of Xianhe was established. The extraction method of tannins of Xianhe was established. The separating condition of flavonids was established by using polyamide colunm chromatography. Screening the sort of macroporous adsorption resin by means of the content of flavonids and triterpenoids, and the SP825 was the best for adsorbing and separating in the following technological conditions: the sample was water liquid, the eluting reagent was 50% ethnanol. And flavonids could be eluted by 50% ethnanol from extract part of n-Butanol as well as triterpenoids could be eluted by 50% ethnanol from extract part of Ethyl acetate. The acute toxicity of Xianhe in mice was investigated and the value of the LD50 of the alcohol extract of Xianhe offering to mouse was counted. To observe the effect of Xianhe, the model of hepatocarcinoma22 solid tumor-bearing mice was produced and respectively treated with the alcohol extract, components, and polysaccharide of Xianhe.
