| Objective: Applying focal ischemia-reperfusion model of SD rat on the condition of hyperglycemia,through observing the state of neurologic impairment score,cerebral pathomorphology,infarction volume,and the expression of inhibitory apoptosis gene bcl-2, intercellular adhesion molecule-1(ICAM-1) and endothelin-1(ET-1),to evaluate the preventive effects of sibelium(SBL) on focal ischemia-reperfusion damage in SD rat on the condition of hyperglycemia.We further compare the preventive effects of SBL with that of bay aspirin (BAY-ASA),to provide some experimental proof for preventive medical therapy.Method:54 healthy male SD rats(weight from 180g to 220g) with normal blood glucemia(<6mmol/l)were randomly divided into 3 groups: hyperglycemia group (n=18), SBL+hyperglycemia group(namely SBL group n=18), BAY-ASA+ hyperglycemia group(namely BAY-ASA group n=18). Each group was divided into 3 subgroups according to reperfusion 3h(n=6),6h (n=6),24h (n=6) after ischemia for 90 minutes. Every animal of SBL group and BAY-ASA group is separately received stomach-douche with SBL of 10mg/kg and BAY-ASA of 6mg/kg once a day for 3 days before ischemia-reperfusion respectively. 50% glucose were injected for all animals in 3g/kg by intraperitoneal injection 30 minutes before ischemic operation to establish hyperglycemia model, blood glucose reach 11.1mmol/L before obstruction means that model is established successfully.Using the improved Longa-Zea method to establish ischemia- reperfusion model in right cerebral middle artery of SD rat, pull out of the... |
