Objective To study the effects of aspirin preconditioning on the production of TNF-α,Caspase-3 in rats focal cerebral ischemia-reperfusion and to explore protective effects of aspirin neuronsMethods 40 Wistar rats were divided into four groups:①Normal control group(n=10)②Sham-operation group(n=10)③Ischemia group(n=10)④Ischemia and aspirin preconditioning group(n=10).To prepare focal cerebral ischemia model, the middle cerebral artery of rats were occluded by a monofilament. The rats were killed at 24 h reperfusion and left rat brain tissue was sampled. Immunohistochemical method were used to investigate the effects of EA on TNF-α,Caspase-3 expression in ischemia area.Results There were no significant difference between normal control group and Sham-operation group. Compared with and normal control group and Sham-operation group, the expression of TNF-a and Caspase-3 were increased obviously in the ischemia group(p<0.05). Among ischemia group, there had some difference between ischemia and aspirin preconditioning group and Ischemia group(p<0.05).Conclusion The application of Asprin preconditioning could inhibit the expression of TNF-a and inhibit the activation of Caspase-3,which might be the mechanism of aspirin achieve neuroprotection.
