Objective To investigate the biological effect of CD40 stimulation on human gastric cancer cell lines AGS and BGC-823 ,and analyze the difference of gene expression profiles of AGS after activating CD40 signalling.Methods Human gastric cancer cell lines AGS and BGC-823 were chosen as target cells and CD40 signalling was stimulated by soluble CD40 ligand (sCD40L). The proliferation of tumor cells was measured by MTT and the flow cytometry. Gene expression profiles of gastric cancer cell line AGS after treatment with 2μg/ml sCD40L for 48h were analyzed by Affymetrix U133A 2.0.Results (1)Inhibition of growth and blockage of the cell cycle were observed in AGS cells with high expression of CD40 after activating CD40 signalling. The number of cells entering G1 stage increased significantly (P<0.05) compared with that in the control group, while the number of cells entering S stage decreased (P < 0.05). But BGC-823 cells with low expression of CD40 didn't respond to it.(2)Both of the gastric cell lines showed no significant changes in apoptosis after activating CD40 signalling...
