Interferential Effect Of Total Glucosides Of Paeony On Mesangial Proliferative Glomerulonephritis In Rats

Objective To observe the influence of the Total Glucosides of Paeony (TGP) on urine protein, serum creatinine(CRE), urea(UREA), total protein(TP) and albumin(Alb) on mesangial proliferative glomerulonephritis(MsPGN) in rats, and to provide the experimental evidence of the possibly protective effect of TGP on MsPGN. Methods The model of MsPGN rats were induced by gastrointestinal comprehensive immunization, I.e. By feeding bovine serum albumin alternate day, injecting Freund's adjuvant into subcutaneous periodically(at day 1^th and 8^th ),and injecting staphyloentero entero-toxin into caudal vein (at day 8^th and 15^th). Than the MsPGN rats were randomly divided into control group, model group, tripterygium glycosides (TPG)-treated group and (high,medium and low dose) TGP- treated group. From 14^th to 18^thweek, Treated group were feeded with TPG or TGP(respectively, TGP: 200,100,50mg/kg^-1 .day^-1,TPG: 16.5mg/ kg^-1? day^-1), Twenty-four h urinary protein contents was examed at different period(week 0^th,2^th,4^th,6^th,10^th,14^th,18^th),In the end of experiment the level of CRE, UREA, TP, Alb were determined, The pathological changes, ultrastructural organization and the immune complex deposits was observed. Results 1. urinary protein: The amount of urinary protein was significantly increased in the model group at 10^th, 14^th and 18^th week (P<0.01, compared with control group), there was significantly difference between 18^th week and 14^th week (P<0.05), After four weeks treatment,the urinary protein was decreased in all the treatement group, the medium TGP-treated group and TPG-treated group was significant (P<0.05, compared with model group); 2. biochemical item: The CRE ,UREA were increased in model group (P<0.01, compared with control group).
