Objective To probe the relationship between microcirculation perfusion and the expression of apoptosis-ralated gene Bcl-2, Bax as well as microvessel density after SAH . Materials and Methods Forty-five adult New-Zealand rabbits of either sex, weighing 1500-2500g were used for this study. All animals were randomly assigned to 3 groups: group A (normal control, n=5), group B (SAH, n=20), group C (Sham-operated control, n=20). Group B and C were randomly subdivided into 4 groups again:1d, 3d, 5d and 7d group, 5 animals per group. The rabbit model of SAH and Sham-operated control was set up by injecting autologous blood or sodium chloride into the cisterna magna (CM). CT perfusion was performed at the 1st, 3rd, 5th and 7th day after the 2nd operation. The CBF, CBV, MTT maps were then obtained, the values were calculated. Group A animals were killed after imaging, group B, C animals were killed at d1, d3, d5, d7 after imaging also. The basilar artery, basal ganglia, temporal lobe cortex were chosen for histopathological examination and morphometric analysis. At last use the samples gathered in each group, the changes of Bcl-2 and Bax protein and CD34 expression in different time after SAH were examined using immunohistochemistry. Results Food intake decrease and the neurological function disorder were found in all animals after SAH.Pathological changes in the basilar artery showing nuclear chromatin of vascular endothelial cell side gathered, and the tight junction between vascular endothelial cells widened; vacuolated smooth muscle cell;...
