Extract of astragalus (EA) is effective components of astragalus which is mainly composed of astragalosides and astragalus polysaccharides. Nuclear factor kappa B(NF-κB) , intercellular adhesion molecule-1 (ICAM-1), tumor necrosis factor-alpha (TNF α ), interleukin-1(IL-1) and interleukin-6 (IL-6) , maybe play important roles in the process of inflammatory reaction during global cerebral ischemia-reperfusion. The project studied the changes of these factors,and observed the expression of MPO reflecting polymorphonuclear cells(PMNs) infiltration by establishing rat globle cerebral ischemia/reperfusion(I/R) injury model. Effects of EA on global cerebral ischemia-reperfusion in rats were studied in this paper. Besides we also investigated probable mechanisms of its neuroprotective effects and to provide some theory for clinical treatment. The main results are showed as follows:1. Effects of EA on global cerebral ischemia and 24h reperfusion in rats This experiment was performed to evaluate effects of EA on acute neuronal injury induced by four vessels occlusion (4VO) for 20min and reperfusion for 24h in rats. During reperfusion after ischemia, EA(40,80mg/kg) significantly improved the recovery of behavioral function.;EA (20,40,80mg/kg) could decrease peripheral blood the amount of WBC. EA (40 mg/kg) could significantly decrease the percent of Non-Lymphocyte and increase the percent of Lymphocyte. EA(20,40,80 mg/kg) could lessen histopathological alterations of cortex in brain sections. EA(20,40,80 mg/kg) might inhibit MPO activity in cortex and hippocampal homogenate. TNF α ., IL-1 and IL-6 content was estimated by radio-immunoassay both in serum and cortex homogenate.EA(40,80 mg/kg) could markedly inhibit the increase of TNF α IL-1 and...
