Astragalus is one of traditional Chinese medicine against cerebral ischemia and the extract of astragalus (EA) is effective components of astragalus which is mainly composed of astragalosides and astragalus polysaccharides. Our earlier research indicated that EA had better protective effects on cerebral ischemia and also had better antioxidation , anti-inflammation and immunoregulation and others pharmacology functions . Middle cerebral artery occlusion(MCAO) was used to make local cerebral ischemia-reperfusion model in rats with intravascular nylon filament occlusion in this paper. Besides we also futher investigated the correlation between the protective effects and its mechanisms of the extract of astragalus (EA) against local cerebral ischemia-reperfusion injury with the anti-inflammation and lmmunoregulation. The main results are showed as follows:1 Protective effects of EA on local cerebral ischemia and reperfusion injury in ratsThis experiment was performed to evaluate the neurological function disorder by Bederson .EA 80mg/kgs EA (40 , 80mg/kg) and EA (20 , 40, 80mg/kg) could respectively decrease the neurological function score for 2s 8 and 24h.EA (20, 40, 80mg/kg ) could reduce the brain edema obviously and lessen histopathological alterations of cortex in brain sections.2 Effects of EA on the inflammatory response and immunoregulation during local cerebral ischemia and reperfusion injury in ratsThis experiment was performed to evaluate effects of EA on local cerebral ischemia injury induced by middle cerebral artery occlusion (MCAO) for 2h and reperfusion for 24h in rats. EA (40, 80mg/kg) could inhibit the increase of circulating endothelial cells number in plasma and also could decrease the counts of WBC in blood; EA (20, 40, 80mg/kg) increased the neutrophil percentage and decreased the T...
