Objective:To observe the expression of Glia fibrillary acidic protein(GFAP) in Muller cells on mild blunt ocular trauma in rabbits and study its relationship with retinal histologic changes and Miiller cells' mechanism of action in retinal trauma. Investigate the expression of GFAP in Muller cells after bFGF treatment, provide theoretical basis to the therapeutical use of bFGF on retinal blunt trauma.Methods:40 rabbits(80 eyes) are randomly divided into four groups:normal control group, contusion group, bFGF therapeutic group, and Nacl control group. The rabbit retina was injured by a 3J energy contusion caused by a free falling iron bar hitting the cornea, then observe the changes of ocular fundus after ocular contusion. The eyes were enucleated at 3h,1h,1d, 3d, 7d, 14d and make it as paraffin sections for HE staining and GFAP immunohistochemistry staining. The morphologic and histologic changes of the retina were observed by light microscopes,in addition, with computer image analysis system,we measured the positive rate and gray scale value of retinal GFAP expression,and analyzed statistically with package of SPSS 12.0 for windows.Results: 1. In the normal control group, the retinal tissue structure is normal,and there is little brown positive GFAP stain in internal limiting memberane. 2.In contusion group and Nacl control group,we can find edema of retinal (especially in the nerve fibre layer and inner plexiform layer) in the early stage after ocular contusion, the retina structure return normal at the later period,but the ganglial cells(RGCs) population decreased. The feature of the retinal histologic changes in bFGF treament group is the follows:the edema of retinal is light compared to the other groups,and the retinal ganglial cells population is also much more than the other groups. 3. The contusion group and Nacl control group GFAP stain begin strengthen at the 3d.reach a hihg lever at the 7d .In bFGF therapy group, GFAP immunolabelling appeared more intense at the 1d after ocular contusion and still rise at the 14d, the...
