| Purpose:To investigate the coding exons in the trabecular meshwork–induced glucocorticoid response protein (MYOC) gene of the patients who had LASIK or LASEK surgery and occurred steroid-induced high ocular pressure and the primary open angle glaucoma patients for mutations, to make a pilot study about the mechanism of these diseases in the gene level.Menthods: Thirty Chinese patients without POAG but who had LASIK or LASEK surgery about one month ago and occurred steroid-induced high ocular pressure after using glucocorticoid according to the rutine were screened for sequence alterations in three coding exons of the MYOC gene by polymerase chain reaction and DNA sequencing. Thirty sporadic unrelated individuals with POAG, aged 50 years or more, were studied as control subjects. At last ,bidirectional sequenced the polymerase chain reaction products and analyzed the sequence alterations include mutations and sequence variations.Results:There were no the sequence alterations in the thirty primary open angle glaucoma patients; but in the patients without POAG but who had LASIK or LASEK surgery about one month ago and occurred steroid-induced high ocular pressure after using steroid according to the rutine, there were two sequence variations. One was mutation , another was only sequence variation because it was not induced the change of the amino acid. In thirty after-operation patients ,we only found 1 patient who had the Arg342Lys mutation in the myoc gene; we discovered 6 patients who had the heterozygosity mutation ,that is 2271 G or A.
