Background and ObjectiveAs one of the etiopathogenisis of tumor, the role of mycotoxin has been identified. It has been observed that mycotoxin can induce mutation of gene and then result in carcinogenesis. And it has been confirmed that Alternariol (AOH) and Alternari-oimonomtyhi ether (AME) have strong carcinogenesis. DNA polymerase beta (DNA polβ)is a member of DNA polymerase family. Its main function is to repair damaged DNA. And it excises and repairs DNA base mutations ranged from 1-4 bases especially. Recently, it has been reported that DNA polβ gene mutations have been discovered in colorectal cancer, esophageal cancer, breast cancer, liver cancer, lung cancer, bladder cancer and prostate cancer. What's more, its expression level in many of them is on the increase. But there is no report on whether it has any relation to the pathogenic mechanism of mycotoxin.In this study, we transfected different plasmids into NIH3T3 cell lines and obtained stable overexpressive NIH3T3 cell lines carrying different plasmids, respectively. We induced the cells with different concentrations of AOH to observe the effect of AOH on the mutation of polymerase β and the change of the protein of the cells. Ultimately, this experiment might provide important evidence for discussing the pathogenic mechanism of mycotoxin in molecular level and the therapy of DNA polβ.
