Objective:The leukemic immunophenotype is using monoclonal antibody (McAB) to detect the expressions of intracellular and nuclear antigens and to analyze the phenotype to obtain the lineage and the differentiation-stage-specific of leukemic cells. The acute leukemic cells are malignant proliferations of hematopoietic precursor cells which are arrestted at immature differentiation stages and expressing the differentiation antigens of the relative blood cells.These antigens can be the markers to detect and classify the leukemic cells.The leukemic cells aren't so the same as the normal ones in antigen expressing.They often lose normal antigens belonging to their developing stages or express additional antigens which lose specificity and differentiation partly.At present the unique antigen of leukemia has not been found.It is necessary to study the rules of the antigen expression in leukemia.Now HLDA has named 393 kinds of CD (Cluster of Differentiation) formally.We often judge the source and differentiation stage of T-ALL, B-ALL and AML by the expressions of CD in leukemic cells .We utilized CD45/SSC gating three-Labelling FCM to analyze the immunophenotype ,diagnosis and prognosis of 310 cases of acute leukemia.
