Primary hepatic carcinoma is one of the most cmmon malignancies in China,comphrehensive treatment is the major mehtod for the inoperative hepatic carcinoma.Chemotherapy has been one of improtant adjuvant treatment in hepatocelluar carcinoma, it plays an importmant role in the comprehensive treatment of hepatocelluar carcinoma.Conventioanl themotheraptic dugs don't often have good therapeutic effects in hepatocelluar carcinoma because of high recurrence rate, low susceptibility and multidrug resistance .so it is important for us to find a new effective chemotheraptic drug..Cyclooxygenase (COX) also known as prostaglandin endoperoxide synthase ,the central enzyme in prostanoid biosynthesis,catalyzes the conversion of arachidonic acid to prostagliandin. It has two distinct isoenzymes COX-1 and COX-2.COX-1 is expressed constitutively in many human tissues and is important for the maintenance of homeostatic functions as the constitutive enzyme. In contrast, COX-2 is a type of inducible enzyme. Under normal condition , it is not detected, but can be induced by mitogen, cytokines or tumor promoters. COX-2 has beend found high expression in colonic, gastric and liver carcinoma and is related to occurrence and development of them in recent years. In addition, lines of evidence have indicated that non-steroidal anti-inï¿¡ammatory drugs(NSAIDs), potent inhibitors of COXs, exert chemopreventive effect on the development of these cancers.Recent studies indicated that overexpression of COX-2 may induce tumor angiogenesis and metastasis.
