In order to searching for the active substances of Egg oil of Rana temporaria chensinensis David (EORTCD). The locomotor activity of mice, convulsion model induced by struchnine and staircase test of mice were introduced to evaluate the biological activity of different products of Rana temporaria chensinensis David. A TLC method and a reversed-phase HPLC (RP-HPLC) method were established in the experiment.Egg of Rana temporaria chensinensis David was extracted by Ethanol, Ether. Acetone, chloroform, Petroleumether and supercritical CO2 extraction. All the extracts reduced the locomotor activity of mice and the number of rearing in the staircase test. The latency of convulsion mice was prolonged. All the extracts showed no difference on the constituent by TLC.The product of supercritical CO2 extraction was further separated by molecular distillation. The products in the 100-160℃ portion showed strong inhibition on locomotor activity, convulsion and anxiety of mice. The effect of the product less than 100℃ on locomotor activity was weaker than on latency of convulsion and anxiety. The part higher than 160℃ was failed to exert an inhibition activity. Combined with the analysis by TLC and RP-HPLC, the result of phannacological experiments indicated that the components with Rf >0.62 on TLC were inert part of EORTCD, while the components with Rf ≤0.62 on TLC were related to the central inhibiting action.
