Objective:This experiment is to explore the mechanism of electroacupunture (EA) regulating hypothalamus-pituitary-ovaries axis (HPO) in the climacteric rats.Methods:Experimental Wistar rats were divided into intact group (INT), climacteric group (CLI) and climacteric with electroacupuncture Zusanli appoint group (CLI+EA). RIA (radio-immunoassay analysis) was used to determine the blood content of E2 (estrogen) , T (testosterone) . The changes of ultrastructure in hypothalamus was observed under the electro-microscopy . RT-PCR used to measured the expression of and GnRHmRNA , NPYmRNA, POMCmRNA and SP mRNA in the hypothalamus of rats .Results:1. Compared with in CLI group, the level of the blood E2 and the expression of Aromatase mRNA in the ovaries increased, but the blood T ,LH, FSH and ACTH.decreased in CLI+EA group.2. The aborration of the cell ultrastructure in the hypothalamus occurred in CLI group. Compared with in CLI group, RT-PCR product of GnRHmRNA, NPYmRNA and SP mRNA in the hypothalamus has decreased in CLI+EA group, but RT-PCR product of POMC mRNA has increased in hypothalamus.Conclusion:The effect of electroacupuncture stimulating ZUSANLI on the function of rats HPO axis on mRNA level may be one of the mechanisms that electroacupuncture cures climacteric syndrome in clinic.
