| Objective: To explore the inhibition of proliferation, differentiation-induction and inducing apoptosis effects of Vernonia anthelmintica Willd. on human melanoma A375 cells in vitro.Methods: (1)The extract Q1 and Q2 were obtained from Vernonia anthelmintica Willd. through following steps: First removing fat, second extracting in Chloroform and Acetone. (2) MTT killing experiment was used to studying the effects of the extract Q1 and Q2 on five tumor cell lines (such as human melanoma cell A375, mice melanoma cell B16, human breast tumor cell BCF-7, human gastric carcinoma cell BGC-823 and human liver carcinoma cell HepG-2). (3) Cell morphology observation, MTT method, HE staining and transmission electromicroscope were used to study the mechanism of the inhibition of A375 cells proliferation by the extract Q2.Results: (1) The percent content of the Vernonia anthelmintica Willd. extract Q1,Q2 were 0.37% and 1.05%. (2) MTT method showed that extracts of Vernonia anthelmintica Willd. had different effects on tumor cells growth, the extract Q2 had significant growth inhibition on human melanoma A375 cells. When the concentration of the extract Q2 was the range from 1ug/ml to 40ug/ml, There is a concentration-dependent relationship between the Vernonia anthelmintica Willd. extract and the inhibition of A375 cells proliferation (P<0.05).
