OBJECTIVETo study the effect of nitric oxide(NO) in the pathgenesis of schistosomiasis liver fibrosis.METHODSTO establish the model of hepatic fibrosis in mice and treated with L-Arg (preNO) and L-NAME(NOS inhibitor). The degree of hepatic fibrosis, the levels of HA, LN, ALT, AST were detected with histological examination, image and chemical analysis, respectively. Change of NO was also measured.1. Murine schistosomal hepatic fibrosis model was built by making the mice transepedermally infected with S. japaonicum cercariae.2. Histochemical stainingThe mice were killed at the end of experience, their liver were cut off and embedded with 4% paraform, then the pathological changes and the deposition of collagen in liver were observed by HE,VG and Sirius red staining.3. The indexes about hepatic fibrosis and hepatic damage were detected.alanine aminotransferase, aspartate, aminotransferase, hexadecenoic, laminin, malondialdehyde, superoxide dismutase, hydroxyproline4. Imaging analysis systemTo explore the extent of liver fibrosis. Volume fractions of collagen were quantitated.RESULTS1. Murine schistosomal hepatic fibrosis model was built successfully.
