| Objective To investigate the polymorphism of IL-1B-511,-1470, the VNTR polymorphism of IL-1RN in Yugu nationality and the difference in the distribution of genetic polymorphism between Yugu and other races. And to address the question whether IL-1B-511,-1470 and IL-1RN were in linkage disequilibrium.Methods Techniques of polymerase chain reaction(PCR) and restriction fragment length polymorphism(RFLP), were used to analyze the polymorphisms of IL-1B-511,-1470 and IL-1RN in 80 Yugu nationality and calculate their genotype frequencies and allele frequencies. The linkage disequilibrium and the haplotype analyses of IL-1B-511,-1470 and IL-1RN were done with the software Arlequin.Results It showed that the allele frequencies of IL-1B-511 were 28. 1% for C and 71. 9% for T respectively, those of IL-1B-1470 were 53. 8% for C and 46.2% for G in Yugu nationality, and those of IL-1RN were 93.8% for I,5.0% for II and 1.2% for IV. I / I genotype was the most common, then I / II genotype. The gene distribution matched the equilibrium law of Hardy-Weinberg. Linkage disequilibrium analyses were revealed that D' and r~2that were the coefficient of linkage disequilibrium neared to 1. The dominant haplotypes comprising the three loci were -511C-1470C-RN I ,-511C-1470G-RN I , -511T-1470C-RN I , -511T-1470G-RN I .Meanwhile, Pearson x~2 test showed P < 0. 05. Conclusion The polymorphism distribution of IL-1B-511,-1470 and IL-1RN had significantly difference in different races or different areas. IL—1B—511 and IL-1B-1470 were in strong linkage disequilibrium. IL-1RN* II was always gone along with IL—1B—511*T. And this study provided the basis for searching the relationship between genetics and diseases significantly. |
