Clinical Significance Of Osteopontin In Patients With Gastric Carcinoma

Objective: To estimate the expression of OPN in the peripheral blood and pathologic tissue from the patients suffered from gastric carcinoma and gastric ulcer, clinical diagnostic and outcome significantion of the serum OPN for gastric carcinoma .Methods: The serum OPN levels were detected by ELISA in 60 preoperative gastric carcinoma patients , 60 gastric ulcer patients and 60 healthy volunteers. The corresponding expression of OPN protein was investigated by immunohistochemistry in surgically resected specimens from 60 patients with gastric carcinoma and bioptic specimens from 30 cases with gastric ulcer.Results: The serum OPN levels were significantly difference among the gastric carcinoma group , the gastric ulcer group and the healthy volunteer group (with a level 114.2dr68.7ng/ml, 67.3±29.8ng/ml, 29.5±17.0ng/ml respectively). Serum OPN in patients with gastric carcinoma were higher than in the other two groups (P<0.01). However, there was no difference in patients with gastric ulcer and healthy volunteers . The OPN levels in gastric carcinoma with or without distant tissue metastasis were respectively 158.7±83.3ng/ml and 85.7±42.0ng/ml, which indicated that the OPN level was associated closely with distant tissue metastasis (P<0.01) . The positive rates of OPN expression were 70% in 60 cases of gastric cancer tissues and 0% in 30 cases of gastric ulcer tissues, with a significant difference correlated to those with lymph node metastasis(P<0.05). The serum level of OPN in the peripheral blood was positively correlated to the expression level of OPN in pathologic tissue. Conclusions :1. The serum level of OPN in the gastric carcinoma group was much higher than that in the gastric ulcer and the normal healthy control groups.2. The expression of OPN in the pathologic tissue from the gastric carcinoma group was significantly higher than that in the gastric ulcer group.3. The serum level of OPN was significantly higher in the gastric carcinoma with distant tissue metastasis than that without distant tissue metastasis,and the expression level of OPN was also significantly higher in the pathologic tissue with lymph node metastasis than that in those patiants without lymph node metastasis.4.The serum level of OPN in the peripheral blood was positively correlated to the expression level of OPN in pathologic tissue.
