| Background: Irritable bowel syndrome(IBS) is a common ,chronic functional disorder, due to the heterogeneity and multiplicity of symptoms ,it is often recurrent in a long term. Due to the IBS which is often induced after the infection of pathogenic bacteria, the correlation between the imbalance of intestinal micro ecology and the incidence of IBS is emphasized. Based on it, consider probiotics drugs like Bifid Triple Viable and Bacillus Licheniformis Capsule are aim to improve intestinal microdysbiosis and also used as therapy drugs on IBS. These probiotic drugs is absent of the direction on IBS, so we have study of some the probiotic strains which have a highly direction on IBS.Objective: To study the treatment effect of the two preparation : combination of clinical isolates of lactobacilli and Clostridium butyricum synbiotics which is composed of the two clinical isolates and inulin on a wistar IBS animal model. To investigate the correlation between probiotics combination, synbiotics to improve IBS symptoms, the alteration of fecal flora and to discuss the potential of mechanism for treatment, so as to offer a new concept for IBS treatment.Methods: The mice were divided into seven groups: model group inulin(prebiotics) group probiotics group(the combination of clinical isolates of lactobacilli and clostridium butyricum; CMS-H002, CMS-H003;) synbiotics group Zelnorm (positive group) antibiotics group(Metronidazole and ciprofloxacin ) and normal group. Except normal group,other groups had intragastric administrated with cold Sodium Chloride (0-4℃) for 14 days to induced a constipation-predominant IBS(C-IBS) animal model. Then, After 14 days treatment, take the general state of mice,24 hours feceal pellets, water content of feces, bacterial flora of feces , colon mucosa for HSP70 immunological detect, and HE coloration pathological detect.Results: 1. The IBS model induced was successfully set up: In the 14th day, except normal group, other groups demonstrated a decent total pellets of one day and water content of feces had not pathological damage. In the 28th day, prebiotics group, probiotics group and synbiotics group all increased in water content of feces, probiotics and synbiotics group improved total pellets of one day,the difference appeared statistical significance (P<0.05)2.The fecal flora was analyzed and indicated. In the 14th day, except the normal group, the quantity of bacteroid increased but the Bacillus bifidus and Bacterium lacticum decreased, which had a difference appeareance statistical significance (P<0.05) .The amount of enterococci, Staphylococcus aureus, Escherichia coli and Clostridium butyricum appeared no difference. After a treatment for 14 days,probiotics group and synbiotics group added the amount of Bacillus bifidus Bacterium lacticum and Clostridium butyricum , the difference appeared statistical significance (P<0.05) .3.There was no difference of HSP-70 expression among the seven groups.Conclusion: 1.The amount of the Bacillus bifidus and Bacterium lacticum decreased possibly relate to the presence of C-IBS.2.The probiotics group and the synbiotics group can increase the amount of the Bacillus bifidus Bacterium lacticum and Clostridium butyricum , both take a treatment effect on C-IBS. |
