Objective: In recent years,Lots of studies shows, renal interstitial fibrosis (RIF) has more relevance to renal function than glomerulosclerosis to that ,so it could show the decline of renal function and prognosis of kidney disease.RIF is the common way to final renal failure and major pathological fundament of various cause injure of kidney disease. The pathogenesis is extremely complicated and it is not very clear.The process involves in cells, extracellular matrix, cytokines, adhesion molecules and their interaction. Transforming growth factor, connective tissue growth factor, hepatocyte growth factor , collagen and others are morely researched at present. Traditional Chinese medicine has a long history in kidney therapy. The benefiting vital energy and promoting blood flow is a kind of method;The shenkang injection(SKI) is a complex traditional Chinese drug and is applicated in chronic renal failure;chronic renal function failure.At present,there are many studies about delaying glomerular sclerosis,but the studies about RFI are limited.in SKI. This study researches the expression of the CTGF,HGF in rats renal interstitium after unilateral ureteral obstruction and dugs intervention.we hope our study can make clear of mechanisms of RIF and offer the experimental basis to clinical therapy.Methods:Healthy male Sprague-Dawley rats weighing 180-220g,bought from Hebei Medical University,were randomly divided into four groups,sham operation group(n=18),operation group(n=18),SKI group(n=18) ,benazepril group(n=18).All the rats were acess to food and clean water ,with the environment temperature 23+2。C and moisture 55+2%.the operation group and therapy group rats were operated with left ureter ligated ,but the sham operation group only free the ureter,not ligaed.The SKI group were received SKI (5g.kg-1.d-1 by daily intraperitoneal injection ),while the operation group and sham operation group were received equal volume water by daily intraperitoneal injection, benazepril group were received benazepril (10mg.kg-1.d-1 by daily gastric gavage ).6 rats from evey group were sacrificed after the surgery ,3 days ,7 days and 14 days.The left renal tissue of all the rats were stainined with HE and MASSON ,so RIF were observed from it with light microscope .Immunohistochemical technique was used to estimate the expression of CTGF,HGF and Col III.The results were analyzed semi-quantitatively with the pathological immage analysis system.The experimental data was demonstraed in mean+standard deviation.The analysis of variance were used to the signifcance test and the relation about the data was measured with linear correlation by SPSS 12.0 , a statistic software.Results: CTGF and Col III expressed much stronger in renal tubule interstitial in the operation group than the other two during two weeks,while two threpy groups distinctly induced the expression of CTGF and Col III (P<0.05,as the operation group ),but two threpy groups have not distinctly distinction (P>0.05). HGF expressed much weaker in renal tubule interstitial in the operation group than the other two during two weeks,while two threpy groups distinctly induced the expression of CTGF and Col III (P<0.05,as the operation group ),but two threpy groups have not distinctly distinction (P>0.05).Left kidney pathology changings as below.The sham operation group rat kidneys were normal like bean from the apperance .The obstrcution kidneys become bigger as time went on.The clear and thransparnet seeper was obstrcuted in the bigger kidneys.The renal cortex become thin,the renal pelvis expanded,minor calyx oppressed and become flat.The histologic observation are shown that the renal tubule expanded,monocyte cells and lymph cells infiltrated in the renal tubule of 3 days ,after the surgery.That change become more clear and renal tubule was shrunck and necrotic 7 days after the surgery,while the inflammatory cells infiltrated less than before and RIF become clear increasingly 14 days after the surgery .The threpy groups could distinctly ameliorae the inflammatory cells infiltration and RIF, but two threpy groups have not distinctly distinction. The sham operation group had no changes like that.Conclusions:Expression of CTGF and Col III is increasingly strong in the renal intertitial of unilateral ureteral obstruction rats and their expression develop more than before as time goes on ,RIF appears. Threpy groups markedly limit expression of CTGF and Col III and inhibit RIF. Expression of HGF is decreasingly strong in the renal intertitial of unilateral ureteral obstruction rats and their expression develop less than before as time goes on ,RIF appears. Threpy groups markedly limits expression of HGF and inhibits RIF, but two threpy groups have not distinctly distinction.
