Renal interstitial fibrosis caused by a variety of reasons that the extracellular matrix components in the renal interstitial excessive deposition. Extracellular matrix components including both normal renal interstitial matrix protein components of the increase and normal existence only when the tubular basement membrane matrix protein deposition. Renal interstitial fibrosis almost all kidney disease progress to end-stage renal failure and a common way to the main pathological basis. In recent years, numerous studies show that the degree of tubulointerstitial fibrosis represented the degree of impaired renal function. Therefore, the search for an effective clinical medicine to prevent renal interstitial fibrosis, delaying renal failure is of great significance.Objects:The experiment used biochemical and pathological morphology, immunohistochemistry, computer image analysis technology and molecular biology, from multi-angle, multi-target to discuss the effectiveness and mechanism of YUSHENKELI. Exploring new theoretical and experimental basis of Chinese medicine and provided new ideas and methods for the future clinical guidance.Material and method:1. Copy rat model of renal interstitial fibrosis and group therapy: forty-eight Wistar rats, half male and half female and 200±20 g body weight. The rats were randomly divided into four groups, namely: the control group, model group, the YUSHENKELI group, Lotensin group. N = 12, respectively, in the course of experimental at 21 d, 28d, 42d three points in time to collect rat kidney tissue and serum specimens.2. Twenty-four hours'urine protein and serum BUN, SCr: quantitative determination of urinary protein. Use automatic biochemical analyzer to measure the level of serum BUN and SCr.3. Renal interstitial fibrosis histological evaluation indicators: the renal lesions using 20% formalin-fixed, paraffin-embedded sections, and through HE, Masson staining to object pathological changes of renal interstitial. 4. Using immunohistochemical methods, semi-quantitative analysis to detect the expression and distribution of HGF, BMP-7, CTGF in renal.5. Using RT-PSCR to detect the relative expression level of CTGFmRNA.Results:1. YUSHENKELI can significantly improve pathological injury of rats, with the protection and restoration of renal tissue, delaying the process of renal interstitial fibrosis.2. YUSHENKELI can enhance expression and distribution of HGF in renal interstitial fibrosis rat, delaying the process of renal interstitial fibrosis.3. YUSHENKELI can enhance expression and distribution of BMP7 in renal interstitial fibrosis rat, delaying the process of renal interstitial fibrosis.4. YUSHENKELI inhibits expression of CTGF and CTGF mRNA. CTGF as the evaluation of renal fibrosis is a good indicator to guide clinical treatment.Conclusions:The experimental results show that YUSHENKELI can enhance expression and distribution of HGF,BMP7, inhibit expression of CTGF and CTGF mRNA. It could delay the process of renal interstitial fibrosis. Therefore, the YUSHENKELI can be more renal multi-level, multi-angle, multi-target play a role in treatment.
