PartⅠ:Objective: To sieving out the hepatoma carcinoma cell line MHCC97-C which has high capability of invasion and metastasis from cell line MHCC97, Investigate the expression of uPA system in hepatoma carcinoma cell and its effect in invasion and metastasis.Methods: Sieving out hepatoma carcinoma cell MHCC97-C from MHCC97 by Transwell cabin. Test the optical delnsity(OD) of two cell lines in 30min,1h and 2h by method of MTT, Test the ability of invasion and metastasis about MHCC97-C and MHCC97 by Transwell cabin. Immunohistochemistry study the expression of uPA and uPAR in MHCC97-C .Results: Compared with MHCC97 and MHCC97-C ,The OD in 30min has no statistical significance (P=0.071); The OD in 1h and 2h has statistical significance(P<0.05).The numbers of invasion and metastasis between this two cell lines was different and the result has statistical significance(P<0.01). The expression of uPA between this two cell lines was different and the result has statistical significance(P<0.01). The expression of uPAR between this two cell lines was different and the result has statistical significance(P<0.01)Conclusions: In vitro, it is feasible to sieving out hepatoma carcinoma cell which has high capability of invasion and metastasis, The uPA system play a important role in invasion and metastasis of hepatoma carcinoma.PartⅡ:Objective: To study the effect of Amilorde which is the synthetic inhibitor of urokinase plasminogen activator to the adherent, the invasion and metastasis of cell line MHCC97 in vitro.Methods: Test the OD of cell line MHCC97-C with 0.2μmol/ml amilorde in 30min,1h and 2h by method of MTT. Determine the inhibitory action of different concentration amilorde to invasion and metastasis of hepatoma carcinoma cell (MHCC97-C) by Transwell cabin.Results: Between control and different experimental group , OD of adherent cells in 30min, 1h and 2h has statistical significance(P<0.01), Meanwhile the quantity of cell which can breakthrough the cell envelope has statistical significance(P<0.01).In the experimental groups , the effect between 0.2μmol/ml and 0.25μmol/ml had not statistical difference(P=0.831).Conclusions: Amilorde can significantly inhibit adherent,invasion and metastasis of hepatoma carcinoma cell(MHCC97-C).In effective concentration ,the inhibitory action stronger as the concentration step up . The concentration of 0.2μmol/ml is the most suitable group.
