| Objective:To study the expressions of the urokinase—type plasminogen activator(uPA)and its special receptor(uPAR) in pituitary adenomas and investigate the relationship with invasiveness of pituitary adenomas.Methods: 61 speciments were classified into invasive group and non-invasive group. The protein expressions of uPA and uPAR were detected by immunofluorescence and Western blot analysis and we use reverse transcription polymerase chain reaction(RT-PCR) to detect their mRNA expressions in all speciments.And we use t test,X2 test and spearman correlation analysis to analysis the different expression and correlation of uPA and uPAR in two group tumors.Results: 31 speciments were invasive pituitary adenomas and 30 spaeciments were non-invasive pituitary adenomas in all detected pituitary adenomas.The positive immunofluorescence result of uPA in invasive and noninvasive pituitary adenomas are 67.74% and 36.67%;the result of uPAR are 58.06% and 33.33%. The PCR result of uPA in invasive and noninvasive pituitary adenomas are 0.5468±0.0464 and 0.4462±0.0305; the result of uPAR are 0.6463±0.0314 and 0.5464±0.0471. The positive Western blot result of uPA in invasive and noninvasive pituitary adenomas are 1.6260±0.1083 and 1.4345±0.0788;the result of uPAR are 1.7273±0.1292 and 1.4322±0.1214.The result showed the expression level of uPA and uPAR much higher in invasive pituitary adenomas than that in non-invasive pituitary adenomas.And there are positive correlation between two markers. The result had statistical significance.Conclusion: The results showed that uPA and uPAR in invasive pituitary adenomas significantly increased. It descripted that invasive pituitary adenomas had stimulated the over expression of uPA and uPAR as for the nature change of tumor compared with noninvasive pituitary adenomas.And this over expression would help tumor cell invade other tissue around it.The result made a prelimilary exploration for our group about degradation of extracelluar matrix and basement menbrance with invasive pituitary adenomas. It also laid the corresponding foundation for Follow-up study. |
