Objective:To observe the inhibitory effect of EGCG on cataractous lenses of rabbits induced by High Glucose.Methods:The rabbits lenses were cultured in vitro and cataractous lenses were induced by High Glucose. After treated with EGCG,the effect of the drug was observed at different time points(48h,72h,96h). Under microscopy, enzymohistochemistry change and structural change of lens epithelial cells and crystalline fiber was measured, the apoptosis of lens epithelial cells was detected using terminal deoxynucleotidyl transferase mediated dUTP-biotin nick end labeling (TUNEL) method, examined the active expression of NF-ΚB in rabbits lens epithelial cells by using immunehistochemis- try technique.Results:1. The opacity and apoptosis in rabbits EGCG group were apparently lower than those of them in High Glucose group (P<0.01). 2. The levels of SOD and CAT were decreased significantly in High Glucose group compared with those of them in control group (P<0.01).The level of MDA was significantly decreased but the levels of other indexes were increased in EGCG groups compared with those of them in High Glucose group.3. TUNEL detection discover,EGCG group each timing apoptosis rate were decreased significantly in High Glucose group (P<0.01). 4. The opacity of the lenses in every group was evaluated. The longer incubated with High Glucose treatment, the more active expression of NF-ΚB, the latter was positively correlate with opaque degree of lens and to a certain extent EGCG had a dose-dependent action in inhibiting the active expression of NF-ΚB a in rabbits lens epithelial cell and relieve opacity of lens.Conclusion:EGCG was an inhibitory role in High Glucose induced rabbit lens epithelial cells apoptosis and could delay and ameliorate the occurrence and development of diabetic cataract.
