Objective: To establish the methods and factors affecting the development of an economical ,practical,stable and reliable animal model suiitable for experimental studies of surgical treatment of carotid atherosclerotic stenosis (CASS). Methods: Using 21 New Zealand rabbits, lesions were created by 3% H2O2 isolated segments of carotid arteries and feeding the rabbits a diet of 2% cholesterol, 6% peanut oil for 4 weeks, 8 weeks and 12 weeks separately. many histological methods,such as hematoxylin staining,special staining and severral kinds of immunohistochemical analyses,were used to illustrate the characteristics of the atherosclerotic lesions, and then to screen out the best method to develop the CASS animal model. Results: The average rates of stenosis were calcuated and discribed as following, 4 weeks group 24.09%±7.81%:8 weeks group 66.22%±21.62%, one blood vesslel of the 8 weeks group emergenced vascular occlusion.;and 12 weeks group: 60.30%±19.94%.The percentages of moderate to severe stenosis of the 8 weeks ang 12 weeks group were all more then 80%.The pathological studys suggested that the atherosclerotic lesions were getting more and more severe as the feeding time went on. Fed the atherogenic diet for 4 weeks, the lesions fell into the fatty streak period of atherosclerosis; fed for 8 weeks, the lesions fell into the mature fibrous plaque period;and fed for the 12 weeks, the lesion were belong to the early atheromatous plaque period. Conclusion: Injured ang fed as described by the authors, the animal model of 8 weeks group is stable and reliable,and it meets the demand of experimental studies of sugical treatments of CASS.
